Riser and PCIe slots in R640 10 Bay Chassis Full Height PCIe Bracket ?!

Based on your enquiry, I can't inform that it is not support, as there is no documented information. As I can see, for backplane of 10X2.5 or 8 x 2.5, the mainboard would be the same. Hence, the installation of riser would be similar. Unless I'm misunderstanding, what I found is that the Full height riser is 2B, whats not clear is how Full height Riser will fit in the Half height slot..

You will need to pair with Riser 1B, as referred Table 4. Riser configurations: 1B + 2B.

What are the part numbers so I can have a look, I'm unable to visualize in my mind as the 10 bay chassis has the 3 half height slots..

PowerEdge R640 has two types of server chassis, one is for 3x Half-Height (LP) cards/brackets and the other one is for 1x Half-Height (LP) + 1x Full-Height (HP) cards/brackets, this means you would have to fully replace the entire chassis or the back section as the front holding drive bays is attached with screws.

On this graphic from the ISM (Installation and service manual) you can see that Riser Card R2A is only a PCIe Board
While this presenting the Riser Card R2B is a whole Riser assembly with its own mounting/cage for the cards.
