Which form factor 7010 do you have?
Only the Mini-tower has 4 SATA ports, SATA0 - SATA3
Desktop and Small Form have 3 SATA ports, SATA 0 - SATA2
Ultra Small Form has 2 SATA ports
And the optical drive, if you have one, probably uses of the SATA ports.

Below is the Optiplex 7010 MT motherboard. Which one is not working? And what device is plugged into that non-working SATA port? List all other SATA devices plugged in and what ports they are using.
24 SATA 3
22 SATA 2
21 SATA 1
17 SATA 0

While in the BIOS, did you go to System Configuration- Drives and make sure all were checked?
The HDD should stay in the blue 17. Are you saying that you tested the optical drive in all three ports =
24 SATA 3
22 SATA 2
21 SATA 1
And when you boot into the BIOS, the optical drive appeared on 22 SATA 2 & 21 SATA 1, but not on
24 SATA 3?

Just a couple of WAGs here...
Do you have a RAID setup? A RAID setup might hide one of the SATA ports.
If you disconnect the HDD that's connected to SATA3 on the motherboard, does that port show up anywhere in BIOS now?

Use a camera and take and post pictures of your BIOS.
* General- System Information- Device Information
* General- Boot Sequence
* System Configuration- SATA Operation
* System Configuration- Drives

Hmm, perhaps the port is just defective on the motherboard. Not sure if it is worth spending the time and money to purchase a replacement motherboard for this 2012 model.

Was SATA3 ever visible in BIOS setup and/or working when a drive was connected to it on this PC?
Have you tried clearing BIOS?
Power off, unplug. Press/hold power button for ~15 sec. Open case and remove motherboard battery. Press/hold power button for ~30 sec. Connect known working drive to SATA3. Reinstall battery (use a fresh battery, if this one is old). Reboot with only mouse, monitor and keyboard attached...

Too bad you didn't report this to Tech Support while it was still under warranty...
I guess you could install a SATA port card in a PCI-e slot, assuming you have an available PCI-e slot, to add a 4th internal SATA port.
