No. It's only one or the other, not both at the same time.
When the Graphics Amplifier is plugged in, the laptop will switch off its internal discrete GPU and use only the GPU in the Amplifier.
When the Graphics Amplifier is unplugged, the laptop will use its internal discrete GPU.
