I have Dell PowerEdge R210 II server with 4Gb of memory installed (one module). And I also have 8Gb memory module. I replaced 4Gb with 8Gb – server is working ok. But mixing memory in different ways got error "Unsupported memory configuration. DIMM mismatch across slots detected".
Can I install them both to get 12Gb of memory?
P.S. From server documentation: "Memory modules of different sizes can be mixed in sockets 1 to 4 (for example, 2 GB and 4 GB), but all populated channels must have identical configurations."

It will support 1, 2 or 4 DIMM's; so your suggested configuration won't work. You can get another 8GB UDIMM to match the one you have to provide 16gb of RAM, one in each channel.

So are you getting the unsupported message when putting ONLY the 8GB DIMM in slot 1? If so, what are the exact specs of the memory you are using?
Provided both DIMM's work in slot 1 without error, you can install both for 12GB, but you will likely get a memory warning if you use slots 1 and 2. To use them both with no warning (although slightly less performance), you would use slots 1 and 4.

Just to verify ... you want slots 1 and 4 (NOT the first and fourth) ... slot 1 is the second slot and slot 4 is the first slot. If you are getting the message with this configuration, then try clearing the NVRAM using the jumpers on the motherboard. If you just get a message whether they are in 1/4 or 1/2 and plan on ignoring the message, you are better off putting them in 1/2.
Also, make sure the BIOS is up to date.

Ha ... my bad ... assumed it was like many of the latest models with 3 channels laid out as 4,1,2,3 :)
As Geoff said, they should be put in slots 1 and 2 (first and third slots - the ones with white release levers) ... see if you get an unsupported message. If you get a message, put them in slots 1 and 3 (first and second slots - this would ensure that all 'populated channels have identical configurations' - only one channel is populated).

I got mine working with 3 slots of 8gb each.
I get the message when booting that the configuration mismatch but it works. I'm running windows 10 64 bits with the 24 GB.
I'm afraid to buy another 8 GB as it differs in brand from the built-in one and continue to get the message.
But as the tests with 24 GB it should work.
My bios is v 2.10.

That would be correct; with a single UDIMM in slot 1. Channel A are slots 1 & 3, Channel B would be 2 & 4. Each channel must match in size and speed.
12GB of RAM is not a supported configuration in this server. 1, 4, 8, 16 and 32 GB are supported only.
