According to the manual, if you want 32GB, then yes, you will need to ditch all existing RAM and get 4x8GB QUAD-RANK (4R) RDIMM's. You can use 1333MHz DIMM's, but they will operate at 800MHz, so don't break your back - or the bank - trying to get 1333MHz memory.
Assuming your new DIMM's are also RDIMM's (RDIMM's and UDIMM's cannot be mixed), either of your proposed configurations would work, although having all three slots per channel (1/3/5, 2/4/6) will clock your memory down to 800MHz, so going with 2x8GB with your 2x4GB will give you 24GB while keeping your memory clock at 1333MHz. If you go with a 6-DIMM configuration, then there is no reason to spend extra money on faster DIMM's since they will only clock down to 800MHz. Also, if you get quad-rank DIMM's (4R), they can be mixed, but the 4R DIMM's must go in the white slots, and your memory will be clocked down to 800MHz ... so, find 1R or 2R memory.
Summary ... get 2x8GB 2R 1333MHz RDIMM's for 24GB total RAM.
Current have RDIMMs in there. I was thinking 24GB might be enough, but may as well go to 32GB. If that is the case I I am pretty sure I need to ditch all the current RAM and go with 4x8GB 800Mhz RDIMMs.
