Modifying one of the u.2 slots is not feasible, the backplanes are in pairs

If you add the kit on the back, you gain 4 slots... is practical if you foretell to need to add more storage in the future.

If you, instead, really need just one extra hdd ( even long term ), and if the front 5.25" bay is not being used, you could buy a 3rd party 5.25" to 3.5" adapter and put there the hdd ( internal cabling would be on you to arrange )

If slot 2 has a nvme ssd inside, means slot 1 + 2 were converted to nvme/U.2 usage.

Slot 1 can accept either another nvme (using an adapter to install it) or an U.2 ssd.

 

Slot 3 and 4 are Sata and can accept any sata drive

2nd question :

On the back of the unit you can install 4 more flexbay slots, it requires a kit

Thank you so much. You are pro. Someone who says that he is my virtual assistant said that I should update my bios. lol, there are a lot of ignorant assistant that they mislead.

I understand that I should buy a HDD box to add new HDD to the slot in other-side. Or I should replace with m2 ssd in slot 2 with the HDD. Is there any reference for that information so I can accept a solution?
