There should not be any issues but I recommend you to backup the container files, the logs, the configurations saved in registry (HKLM\Software\OTG key), the \Program Files\OTG\EmailXtender folder first before you do the OS upgrade. There should not be any issues, just that you will need to setup message queuing and application server in the EX server.
I myself haven't done any upgrades (just reinstalled on the windows 2003 server) but I should let you know what you should prepare yourself well before doing this upgrade because u will need to get those container files back.
Before doing the upgrade, don't forget to close all the open volumes so that the files are written to the disk and you can back those up.
I would also do a test if possible to see if there are any issues that come up.
