I would really appreciate if you could test it out for me if it's not too much trouble. Buying 2 8gb sticks isn't an option for me right now, but I'd really like to get above 8gb and not end up with an extra 4gb stick that I'll probably end up getting rid of eventually.

@RalphParker
Pulled an E5540 from the warehouse for testing.
Used 4GB and 8GB memory:
Total of 12GB memory:
Booted to BIOS and started to load W10.
12GB works just fine.
Best regards,
U2

Welcome to the Dell Community @RalphParker
12GB is not listed as a valid configuration.
In fact all I can find is that the max is 16GB
Table 3. MemoryFeature Specification
https://www.dell.com/support/manuals/us/en/04/latitude-e5540-laptop/delllatitudee5540_om/specifications?guid=guid-fa08d88f-67d7-4277-a7b9-d872f106a82b&lang=en-us
I can pull an E5540 from my warehouse in the morning and test it for you.
But remember to take advantage of "Dual-Channel Memory" you must have matched pairs!!!
https://www.crucial.com/usa/en/what-is-dual-channel-memory
That's why most people will purchase matched pairs.
