It is not recommended to hot swap cables every time.  For troubleshooting, it does work for me at least.  The use of adapters could cause issue as well.  Note that signals can be uni-directional (one direction) from DisplayPort (PC) to HDMI (monitor).  Some high quality DisplayPort to HDMI cable (not adapter) can work well.

Many users have utilized the optional DisplayPort in combine with integrated DisplayPort for multi-displays setup.  They all should be working, so double check your display setup again.  Testing with one connection at a time.  

As for daisychaining, DisplayPort 1.2 and higher supports MST.  Verify that your monitors are MST supported, then you are ready for the setup.

@Chino de Oro​ If I exchange the cables from the two monitors they both work but only when plugged into the ''optional'' port, not the 2 other display ports. Windows did not detect a second monitor....Update!!! I tried the other port and it seems to work! Perhaps it does output to second port if there is nothing on first port?

if you want to connect two monitors via Display Port cable to pc, it may be easier to uninstall the optional video module, and simply connect both monitors to the permanently built in DP ports at back.

Both of the DisplayPort on OptiPlex systems should work and when adding the optional port, all three should work.  You could find confirmation from others whom were adding optional port as alternative of discrete graphics for their multi displays setup.  In some cases, you can connect second monitor after the system already boot up.

At this point, it seems that you managed to get signals from one integrate DisplayPort and one optional DisplayPort to work for your dual monitors setup?

@Chino de Oro​ yes this is correct but the connection on the integrated port is sensitive to touch. Is it OK to hot swap monitor cables through these ports? It would seem that audio may not be passed through both ports at the same time but this may be bad contacts. I am using adaptors from hdmi cables. Thank you for your assistance!
