Vault drives do not use Hot Spare unless you have created a Pool using the vault disks. If you do not use the Vault drives for user data (not configured in a Pool), then there is no Hot Spare requirement. When a Vault Drive fails, you just replace the drive (Vault drives can only be physically in the first four slots).
OK, according your answer, i assume that i need a spare drive for the vault disks?
Because in the VNX2 the spare policy alert me if i don't have the necessary disks to spare the first four disks (vault drives), assuming that those ones are a different type compared with the data disks, used in storage pools.

besides a 1.2 TB SAS disk can spare for a 600 GB SAS disk
please see the hot sparing rules in the white paper and FAQ

VNX2 uses the same sparing rules as Unity - only enforcement is different
please re-read the rules

while the system area itself on the "vault drives" doesn't need a spare any user data on it does

in terms of computing how many spares are needed the number of total disks are used - and that includes all disks installed in the system
it doesn't matter for the spare count needed whether or not there is user data on the system drives

Keep in mind that the hot spare quantity has slightly changed in the just released Unity SP1
It changes from 1 spare for 30 drives to 1 spare for 31 drives.
