No, each processor must be populated with the same configuration. So, you CAN'T use a single 16GB DIMM in this server with two processors ... each processor would have to have a 16GB DIMM.

You will always get an error installing only one 16GB DIMM. Do you have one or two processors installed?

The main rules for memory installation on servers using (modern) Xeon processors:
- each memory channel needs to be populated the exact same way, unless a memory channel is not used at all
- when using multiple processors, all processors need to have the exact same memory population
So, you will need to buy at least 1 more 16GB dimm. Then you could do this:
CPU 1: Memory channel 1, slot 1: 16GB, slot 2: 4GB, slot 3: 4GB
CPU 2: Memory channel 1, slot 1: 16GB, slot 2: 4GB, slot 3: 4GB
To upgrade to more in the future after this, you'll have to either remove memory or buy at least 2 x 16GB + 4 x 4GB each time.
