VMware's filesystem is cluster aware - so all hosts can access the same lun at the same time.
you are talking about Windows 2016 - do you plan to use ist directly on the blade or will this be a Windows VM inside VMware. If you plan a VM - you do not need to modify the config, because VMware will handle the access to the LUNs.
If you plan to go physical with Windows 2016 - it must not access the luns advertised to VMware!
