computer to computer
I have a new netbook which, of course, has no optical drive in it. Can I connect my desktop and netbook with a USB cord and use the DVD drive in in my desktop to upload/download files to the netbook? I don't want to buy an external drive because I'm reluctant to spend $50-75 on something I'd use only 2-3 times a year.

Not as such, no. You CAN connect the two systems with a pair of Ethernet cables and a hub or switch, or a crossover Ethernet cable - or a USB networking adapter. USB to USB can't be done with out such a device between the systems.
