Spybot, and maybe other ad blockers/malware stoppers, can lock IE's settings, giving symptoms like what you described. Are you running Spybot with "lock IE settings" selected?
I sounds like you have acquired a homepage hijacker. This is likely the cause of your other problems that you posted in your other message as well. Go to the following page and first download, install, update, and run Spybot S&D. After you let spybot get rid of spyware it finds, download and run cwshreddrer. Then, if the problem is still present, download and run HijackThis and post the log file that it creates here for further suggestions.
