You should get an A:\ prompt when booting to a startup diskette. If you're trying to access the C: drive, at the A:\ prompt, type the following:
cd C:\ (press Enter, which should take you to another A:\ prompt)
type C:\ and press Enter. You should now be at the C:\ drive, and can do whatever you need to do with it from that point.
If, on the other hand, you're trying to reinstall Windows from a CD, etc., you're going to have to be more specific about what it is you're trying to do, as it isn't evident in your first post. For example, if you want to reinstall from a CD, you should first FDisk and format the hard drive, using the boot/startup diskette, then reboot to the startup diskette, and at that point, one method of reinstalling from the CD would be to substitute your CD-ROM drive letter for the C:\ above, and run Setup from there.
Example (assumes drive has been fdisked and formatted)
Boot to the WinME startup diskette, which should eventually get you to an A:\ prompt.
type cd E:\ (use the appropriate drive letter for your CD-ROM drive - may be D:. etc.) and press Enter. You should get another A:\ prompt. Type E:\ and press Enter, which should get you to the prompt for your CD-ROM drive. At this point, type setup and press Enter, which should start Windows Setup from the CD-ROM.
Example:
E:\setup (assumes OS CD is in CD-ROM drive) - press Enter. (Windows Setup should now commence.)
