The big difference is in the support you get. You can only buy a Latitude from the business section/area of Dells site and with that you get a warranty that is more business oriented. Like real tech support and next business day service when you buy the extended, onsite and complete care warranty. Other than that there really is no difference between them other than some external parts being slightly different.
The Dell Latitudes come standard with a 3 year warranty while the Inspirons typically come with the skimpy 1 year warranty. Unless this has changed. The Latitude line also has additional security features.
The Latitude line uses better materials (i.e. magnessium) in their construction while the Inspirons employs mostly plastics.
