I did an upgrade on my E510 and worked fine.  I went from the Dell OEM Media Center edition to the Retail Vista Home Premium update. 
 
However, I had downloaded all the Vista drivers I needed before doing the upgrade and also ran the Vista readiness tests - both the Microsoft and the Dell and made sure everything was OK.  The Dell flagged the BIOS and after upgrading to the latest BIOS for the E510 the Dell readiness test was "happy".

absolutely, I got fed up waiting for my Dell upgrade and purchased the Ultimate version. Upgrade the Dell drivers and installed all working fine

And you get the 64 bit version as well.  Dell does not do this at this time.  Unfortunately you had to fork over the extra money, and that is a shame
